高效甘薯脱毒苗生产及驯化移栽

摘 要:

选用目前主要推广的甘薯品种‘烟薯25’、‘商薯19’和‘徐薯22’为材料, 研究了甘薯脱毒快速繁殖及驯化移栽。结果表明, 秋天刚收获的薯块在36 ℃条件下催芽, 10~15 d后芽苗可长到10 cm以上; 剥取带有1~2个叶原基的茎尖进行培养, 3个供试品种植株再生率均达到90%以上; 脱毒试管苗炼苗2~3 d后直接栽植于塑料大棚, 移栽成活率达95%以上; 移栽5~6周后茎蔓可长到30~40 cm以上, 即可剪切茎蔓扦插进行再繁殖。

Production, Acclimatization and Transplantation of Virus-Free Plantlets of Sweet Potato

Abstract:

The method on production, acclimatization and transplantation of virus-free plantlets was studied by using cultivars ‘Yanshu 25’, ‘ Shangshu 19’ and ‘Xushu 22’ of sweet potato (Ipomoea batatas). The results indicated that the seedlings could grow more than 10 cm after a germination acceleration at 36 ℃ for 10–15 d. Then, stem tips with 1–2 leaf primordia were excised and cultured on the induction medium, the plantlet regeneration rates were above 90% in the three cultivars. The virus-free plantlets in tubes were acclimated for 2–3 d and subsequently transferred to soil in plastic greenhouse, and the transplanting survival rates of the three cultivars were over 95%. After having been transplanted for 5–6 weeks, the lateral branches reached 30–40 cm in length, and they can be used for propagation in plastic greenhouse.
